About 4 days, like Jewelrose said. You don't have to be there while she has pups and her mood will turn to "Labour" while she is giving birth. Labour will last a few hours.

I had 4-5 wolfs that were pregnant and after they all gave birth i only had 2 puppies out of the 4-5 that were pregnant. Where did they go?

If you didn't have enough room the pups would have wondered off. So any pups that were going to be born weren't generated as there wasn't enough den space with in your den for you to keep them.
