I think one thing that helps me with my characters is knowing what drives them. One of my characters is an assassin, but his whole goal in life is to stay alive. Not much of a goal, but it helps me determine how he is going to act. Of course that might not help make them more interesting, but it helps me keep a grasp over who they are and what they are like. I feel like some of my characters are sort of just there, and I don't know what to do with them or how to act, because I don't know their governing movtives

Mannerisms and quirks always spice up a character a little bit, I think. Just try not to overdo it or it may become irritating. Another thing that may make your character unique is an uncommon attitude towards common human experiences. I mean things like death, love, family, money, all that. If you give them a unique attitude towards one of these, people will notice. If you're doing an RP set in another world, you can either make up your own culture (only if the owner says it's okay) or entrench your character deeply in one of the pre-existing cultures.
