I fill the lines with a mid gray, and then shade them using a brush called 'digital brush' in Autodesk Sketchbook. Then I blend it out using the blur brushes. I repeat the process with darker grays until I'm happy with it, then put in a few highlights with a light gray.

Then I make a layer above the shading layer, set it to 'overlay' and colour in the characters markings on that layer so the shading appears through :)
