Yes, you can breed a wolf with the Infatuation boost anytime. You have to wait until the next wolf party event for a first generation one, though

Is the breed quest supposed to be fulfilled immediately once you've bred a suitable pup, or do you have to wait for rollover or something?

I've had a brindle pup quest since I joined and one of my wolves had a brindle pup a few hours ago, but the quest is still there.

What was the exact quest? Maybe it was more specific and brindle was only part of it?

It's says breed a brindle pup and nothing else as far as I can see

Could be a bug, maybe try to get another bindle pup and see, if not report as a bug
